JP: 顕微鏡の試料台上に高品質の白色リファレンスを置くことはできますが、顕微鏡を反射(エピ)照明モードにする必要があります。これは、対物レンズから試料に光を当てていただくこととなります。
これが適切に設定されれば、標準的なホワイトの校正と同じです。
反射型顕微鏡は、透過型や他の方式に比べてS/Nが悪くなりがちなので、あまり使用しない傾向にあります。この方法は、低倍率で高照度の光源を持つ工業用検査画像顕微鏡に適しています。
EN: One can place a high quality white reference on the microscope specimen stage but it means the microscope will need to be in reflectance (epi) illumination mode. That is light has to be directed through the objective lens onto the sample. If that is properly set up then the rest of the process is the same as the standard white cube calibration. We have found that reflectance microscopy tends to yield low signal to noise levels compare to transmission and other methods so tend not to use it. This approach is more appropriate for industrial inspection imaging microscopes with low magnification and high illumination sources.
